PhD Candidate for Development of Novel Immunometabolic Platforms for Cancer Diagnostics and Pharmacotherapy

PhD candidate for Development of novel immunometabolic platforms for cancer diagnostics and pharmacotherapy (1.0 FTE)

Project description

We will develop in vitro technologies to probe tumor immunometabolism at the single cell resolution. The technology will allow us to characterize tumor associated macrophage heterogeneity, study the role of the microenvironment on their function, and to monitor their behavior upon administration of inhibitors of metabolism. This could lead to a breakthrough in cancer immunotherapy and diagnostics. Key responsibilities

The aim of your PhD project is to develop an advanced analytical and in-vitro platform for metabolic phenotyping of tumor associated macrophages. You will establish miniaturized metabolomic setups to profile metabolites for immuno metabolic characterization of 100's of cells in cancer relevant microenvironments down to individual cells. You will optimize sample preparation in such a way that the extract after sample preparation is ideally suited for CE-MS and nano/micro LC-MS analysis. You will develop a robotic sample preparation platform and optimize nano LC-MS protocols. You will optimize sampling from an innovative cancer-on-a-chip platform, which you will help to further develop. You will work with industrial partners (Agios, Mimetas, Sciex, SampleQ) to learn about relevant modules for this project and to test novel mass spectrometry systems and prototypes, and novel organ-on-a-chip models. Within the LACDR there is a growing focus on using (translational) metabolomics and patient derived organ-on-a-chip systems in the areas of drug research, translational disease research, systems pharmacology, computational modeling and biostatistics, and integration with epidemiological studies, genetics and clinical studies. You will work closely with the Leiden Centre for Data Science.

The Analytical Biosciences and Metabolomics group at the LACDR has a world-leading reputation in the field of metabolomics, and has a track record in developing novel analytical and microfluidic methods.

Terms and conditions

We offer a position for initially one year. After a positive evaluation of the progress of the thesis, personal capabilities and compatibility the appointment will be extended by a further three years. Salary rangefrom 2,325 to 2,972 gross per month (pay scale P in accordance with the Collective Labour Agreement for Dutch Universities). Mobility and family allowances as applicable according to Marie Skodowska-Curie PhD fellowship scheme for early career researchers.
